 A large PDZ containing family of actin-binding proteins concentrated at the adherens junction (Ikeda et al, 1999). Afadins bind a variety of proteins at the junctions.  One of the afadin binding partners are the nectin family of adhesion molecules (Reymond et al, 2001).  The nectins are homologs of the group of proteins that include the poliovirus receptors (PVR/CD155/PRR).  Afadin is similar to AFAP-110 in that it binds actin through a C-terminal domain but the actin-binding region (1631-1829) is not obviously similar to other actin-binding motifs (Mandai et al, 1997).
